English: Transcriptional activation of E3 (top) and E6 (bottom) cells. The DNA locus is tagged with CFP-LacI and the cytoplasmic peroxisomes are also seen in CFP. The mRNA is tagged with YFP-MS2. As transcriptional induction proceeds, the transcription sites appear and then mRNAs disperse into the nucleoplasm. CFP-peroxisomes appear at later times. Since the videos were acquired in 3D and the z-stacks covered the cell volumes, we could show the nuclear CFP-LacI locus in the beginning of the videos and then later shift to the cytoplasmic plane that best shows the CFP-peroxisomes. Cells were imaged 15 min after dox addition and the time in the videos is measured from dox addition. E3, imaged every 15 min; E6, imaged every 20 min. |
