Українська: Імунофлуоресцентне забарвлення сірої речовини поперекового відділу спинного мозку щура у фіксованому препараті, результат комбінації серії конфокальних зображень (мікроскоп Olympus FV 1000, об'єктив 60x NA 0.9, водяна імерсія). Зеленим кольором представлено важкі ланцюги нейрофіламентів (neurofilament heavy chain - NfH, антитіла до NfH із барвником AlexaFluor 488), компоненту проміжних філаментів нейронів. Блакитний колір представляє забарвлення ДНК у ядрах гліальних клітин (Hoechst, флуоресцентний барвник специфічний до ДНК), візуальна відсутність забарвлення ядер нейронів пов'язана з обмеженнями динамічного діапазону мікроскопа, інтенсивність флуоресценції ядер нейронів була на декілька порядків нижча за інтенсивність ядер гліальних клітин. Пурпуровий колір - відбиття довжини хвилі збудження 488 нм від щільних компонентів позаклітинного матриксу, зображення отримано шляхом використання фільтра збудження 488 нм із реєстрацією в діапазоні 488-505 нм, отриманий сигнал значно перевищував інтенсивність флуоресценції барвника AlexaFluor 488. Філаментозна структура на флуоресцентному зображені збігалася зі спостереженнями з використанням диференційно-інтерференційного контрасту. Також не спостерігалося зниження інтенсивності отриманого сигналу через фоторуйнацію з чого було зроблено висновок про природу утворення зображення шляхом пропускання частини відбитого від зразка лазерного випромінювання 488 нм через недовершеність оптичного фільтра збудження. English: Immunofluorescent staining of the gray matter in the rat spinal cord cross-section in a fixed specimen, resulting from a series of confocal images (Olympus FV 1000 microscope). Neurofilament heavy chains (NfH) are depicted in green, stained with AlexaFluor 488-labeled antibodies. Neurofilament heavy chains are components of neuronal intermediate filaments. Blue color represents DNA staining in glial cell nuclei using Hoechst, a fluorescent dye specific to DNA. The absence of neuronal nuclear staining is due to limitations in the microscope's dynamic range, with neuronal nuclear fluorescence intensity being several orders of magnitude lower than that of glial cell nuclei. The purple color indicates the reflection of the 488 nm excitation wavelength from dense extracellular matrix components, imaged using a 488 nm excitation filter with detection in the 488-505 nm range. The signal obtained significantly exceeded the intensity of the AlexaFluor 488 dye fluorescence. The filamentous structure in the fluorescent image correlated with observations using differential interference contrast. There was no observed reduction in signal intensity due to photobleaching, leading to the conclusion about the image formation through the passage of a portion of the 488 nm laser emission reflected from the sample through the imperfection of the excitation filter. |
