(A, B) Longitudinal semi-thin sections (1 μm) depict the columnar arrangement of proliferating (pc) and hypertrophic chondrocytes (hc) and the underlying primary ossification centre (poc). (A) At the chondro-osseous junction (highlighted in yellow), osteoblasts (arrowheads) line the mineralized cartilage scaffold and in (B) capillaries (arrowheads) invade the lacunae of late hypertrophic chondrocytes. Toluidine blue stain. (C) TRAP staining. Chondroclasts (red staining) erode the mineralized cartilage and apparently allow the invasion of vessels into the lacunae. Arrowheads point to osteoblasts. The section is counterstained with haematoxylin. (D) Several late hypertrophic chondrocytes contain a shrunken nucleus (n) with condensed chromatin on its margin, rough endoplasmic reticulum (rer) and large amounts of electron dense material, presumable glycogen. The depicted chondrocyte is slightly detached from the cartilage matrix and does not contain any vacuoles. The ultrastructural features indicate that this cell undergoes apoptosis but not autophagy. All sections are from a two-week old mouse, and the bones are demineralized.
