Description
English: Time-lapse imaging of U87MG cells transfected with HttQ103YFP-pIRES-mRFPu pre-treated with SKF86002 two hours prior to transfection. The movie shows a decrease in IB formation and UPS dysfunction. Live cell imaging was initiated at 24 hours post-transfection, with images acquired every 10 minutes. Cells were visualized under white light, and filters that detect YFP or RFP. The brightfield and fluorescence emanating from the YFP and RFP channels were merged to create a movie file. Images were acquired using a 10× objective for a total of 24 hours. Throughout the movie there are fewer cells with IBs (relative to untreated cells in movies S1 and S2). The levels of the mRFPu reporter protein (red colour) remain lower than in untreated cells and do not increase until late in the movie.
