English: mRFP-Toca-1/GFP-N-WASP induced tubule/vesicle transitions with TIRFM. CHO cells were transfected with cDNA encoding mRFP-Toca-1 and GFP-N-WASP. After 24 hr, the time-lapse acquisition was performed on an inverted Olympus FV1000 TIRF microscope at 10 sec interval for 60 min. The penetration depth was approx 100 nm from the coverslip. Image acquisition in the mRFP and GFP channels was taken sequentially. Bar = 10 µm. |
