- The colours arise because of the birefringence exhibited by the crystalline film. When a sample possessing birefringence in its crystalline structure is viewed in between two crossed-polarisers, you will see beautiful colours. The colours depend on several factors including the thickness at each point and the orientation of the crystal axis with respect to the optic axis of the microscope. When a wave plate is inserted in the optical path of the microscope, it introduces a phase shift in the light beams, ultimately producing colour. Different waveplate thicknesses can give different colour palettes. Colour is not artificially applied, it's derived by the physics and chemistry of the sample and setup.
