Bafetinib chemical structures
[edit]There seems to be some inconsistency in chemical databases in relation to the chemical structure of the drug candidate bafetinib (also known as INNO-406). There is a discrepancy in the connection between the two pyrimidine rings. PubChem, ChemSpider, and World Health Organization all list the structure as File:Bafetinib 2D skeletal.svg which corresponds to CAS#887650-05-7. However, Chemical Abstracts lists bafetinib and INNO-406 as synonyms for CAS#859212-16-1 which corresponds to the structure of the three files listed above. I am inclined to think that Chemical Abstracts has the incorrect structure and that the WHO source, which is based on regulatory submissions from the company developing the drug, is the authoritative document in this case. - Is anybody able to find out something about the synthesis of the compound? Maybe a publication covering this will bring some light to the darkness...--Mabschaaf (talk) 16:36, 12 March 2013 (UTC)
- This journal article and the European patent EP1702917 both describe the synthesis of CAS#859212-16-1. I cannot find any publication for the synthesis of CAS#887650-05-7. This would suggest that Chemical Abstracts is right and PubChem/ChemSpider/WHO are wrong, contradicting my argument above. Ed (Edgar181) 17:29, 12 March 2013 (UTC)
- Analogy with the related drugs w:en:imatinib and w:en:nilotinib also suggest CAS#859212-16-1. Ed (Edgar181) 17:32, 12 March 2013 (UTC)
- doi:10.1182/blood-2005-06-2209, authored by the team who discovered/developed the drug, also has the 859212-16-1 structure. It appears WHO (and, consequently, my File:Bafetinib 2D skeletal.svg) got it wrong. Yikes. Fvasconcellos (t·c) 18:39, 12 March 2013 (UTC)
Withdrawn: It's not clear which of the two structures is correct, and in fact it seems more likely that the three files I nominated are the correct one. Ed (Edgar181) 11:57, 18 March 2013 (UTC)
What about renaming the structure(s) that are likely not show bafetinib correctly? If the CAS number is certain, it might be used in the title. --Leyo 22:54, 19 March 2013 (UTC)
- It's probably best to wait until there is a definitive answer about the correct chemical structure. Ed (Edgar181) 12:29, 21 March 2013 (UTC)
- If we are talking about a few days, I agree. On a long term, we should not have incorrectly named structures without at least a comment concerning the doubts in the description page. --Leyo 13:12, 21 March 2013 (UTC)
- Yes, it might be helpful to leave a link to this discussion on the files' talk pages or in the files' descriptions. Ed (Edgar181) 13:21, 21 March 2013 (UTC)
- I added a comment to File:Bafetinib 2D skeletal.svg. The talk page is not visible enough for readers. --Leyo 14:25, 21 March 2013 (UTC)
