Summary

[edit]
Description
English: Many drugs fail at clinical stages, either because they are ineffective or they are toxic. It is also because of the inefficient preclinical target verification. Therefore, target verification is a critical step in drug discovery and development. The purpose of the target verification is to prove that biological targets play a key role in the disease process, and the regulation of the target itself can play a therapeutic role without toxicity to normal cells and tissues. Finding a selective molecule for proof-of-concept pharmacological studies can be achieved promptly. The early target validation process requires multiple efforts, ranging from genetic manipulation of the target in relevant cells, to interrogation of the consequence of target manipulation on pathways, as well as the omics analysis in proper cell systems or in vivo models.
