Since both the main MCU and each channel controller run on quartz crystals the communication almost certainly uses UART.
All optocouplers seem to be connected in series and parallel, respectively (and the mezzanine header does not have enough pins anyway), so each direction is broadcast, which means the individual channel MCUs have to know their channel number, either through firmware or pin strapping.
